Nanocrystal Technology For Oral Delivery of Poorly Water-Soluble Drugs

چکیده
Nanotechnology has been improving and has a variety of application fields. One of the major application areas of nanotechnology in pharmacy is nanoparticular drug delivery systems. Preparation of drug nanocrystals to improve the solubility of poorly water-soluble drugs for oral delivery is also one of the important applications. Nanocrystal dispersions comprise water, active drug substance and a stabilizer. They are physically stable due to the presence of stabilizers that prevent reagragregation of the active drug substance. Different techniques can be used to prepare nanocrystal formulations of a drug powder such as homogenization, coprecipitation, spray drying and milling. There are several advantages of nanocrystal formulations such as, enhanced oral bioavailability, improved dose proportionality, reduced food effects, suitability for administration by all routes and possibility of sterile filtration due to decreased particle size range. The first nanocrystal drug product, Rapamune® (Sirolimus), was approved by the FDA in 2000. In the following years, several other nanocrystal drug products came out on the pharmaceutical market. Nanocrystal technology is cheap and easy to apply; thus, it appears that this technology will be substantially useful for the manufacture of poorly water-soluble drug products for oral delivery.

Nanosized Technological Approaches for the Delivery of Poorly Water Soluble Drugs
A major hurdle in pharmaceutical formulation is water insolubility of most of drugs affecting their stability and bioavailability. If the drug is also insoluble in organic medium, it is difficult to deliver it in a sufficiently bioavailable form and hence it is a great challenge to formulation researchers to overcome such difficulty. Although some approaches are available for enhancing th...

Due to low cost, ease of administration, and lack need for trained personnel, the oral route is the most convenient and accessible way to design different medicines that could be simply consumed by patients. Regardless of the great benefits of this route, the main challenge in the bioavailability of oral medications is gastrointestinal instability. Nanotechnology is used to improve the solubili...
متن کاملEffects of Spray Drying and Spray Chilling on Ibuprofen Dissolution
The formulation of hydrophobic drugs for oral drug delivery is challenging due to poor solubility, poor dissolution and poor wetting of these drugs. Consequently, the aim of this study was to improve the dissolution of a model poorly water soluble drug, ibuprofen. Microparticles containing ibuprofen were produced by spray drying and spray chilling technology in the absence/presence of a ...

Of all the pharmaceutical compounds, about 40% are considered poorly water soluble and hence, the development of efficacious formulation is being halted. It has been shown that a drug should have a minimum aqueous solubility of 1% to avoid bioavailability problems. During preformulation study, sufficient time should be invested in order to improve the solubility of these kinds of drug. At the v...
